Nicotinamide mononucleotide ("NMN" and“ β- NMN ") is a nucleotide derived from ribose, Nicotinamide, Nicotinamide nucleoside and nicotinic acid. In humans, many enzymes use NMN to produce Nicotinamide adenine dinucleotide (NADH). In mice, it was proposed that NMN was absorbed in the small intestine within 10 minutes after oral administration and converted into Nicotinamide adenine dinucleotide (NAD+) through the Slc12a8 transporter. However, this observation has been questioned and the matter remains unresolved.
Due to NADH being a cofactor of mitochondrial internal processes, sirtuins, and PARP, NMN has been studied as a potential neuroprotective and anti-aging agent in animal models. When NAD+levels increase, inhibiting mitochondrial decay can reverse aging at the cellular level, making it widely popular in anti-aging products. Dietary supplement companies actively promote NMN products, claiming to have these benefits. However, so far, there has been no human study to correctly prove its anti-aging effect. A study by Keio University shows that a single dose of up to 500 mg is safe for men. In 2021, a clinical trial found that NMN improved the muscle insulin sensitivity of pre diabetes women, while another clinical trial found that it improved the aerobic capacity of amateur runners.
